OverviewProSpectrum Libraries™ are used to prepare samples for proteomic investigations from a broad range of complex biological materials that are compatible with ProSpectrum Libraries™, including anti-coagulated whole blood, serum, bacterial extracts, tissue homogenates, and cell culture media. A major limitation to evaluation of biological samples is the inability of current techniques to detect trace analytes in the presence of abundant proteins. ProSpectrum Libraries™ overcome this limitation by collecting the protein species present in the original sample and bringing them within concentration ranges that can be more easily analyzed by standard methods. The technology uses millions of peptide ligands synthesized on microscopic chromatography resin beads. These beads use affinity interactions to isolate unique proteins, enrich for trace proteins, and deplete abundant species. In addition, immobilization of the proteins on the library improves their stability and thereby their detection. ProSpectrum Libraries™ also decrease artifacts associated with blood product sample preparation, because whole blood can be incubated with library immediately upon collection. In other methods of sample preparation, such as immunodepletion, the target proteins are diluted as they flow through the column. In contrast, trace analytes are concentrated on ProSpectrum Libraries™. There is no limit to the amount of sample that can be applied to the library; therefore sample can be applied until the library captures enough target protein for detection.
